The gym is a place where you can enhance your cardiovascular endurance and stimulate muscle growth, but any body composition changes are going to be pretty much completely determined by diet.
Burning calories at the gym can help but having the willpower to eat right is the most important part about losing weight. You can run for an hour and loose a few hundred calories but just one more mcDonalds sundae will make it all for naught. You need to have the strength to not eat the extra icecream and get over the calorie threshold for losing weight.

Doing cardio with that much excessive weight will do your joints no favor. If you still want to do cardio then swimming and the stationary bikes are the best options. The treadmill and the stair master will reck your joints for sure.
